The Bird and the Bee, Imogen Heap, St. Vincent, Santigold, Lykke Li. And this is more on the folky side, but Laura Marling has a beautiful voice.

- Skin of Skunk Anansie
- kaRin of Collide
Oh and I second the Siouxie Sioux of Siouxie and the Banshees suggestion above.
Most of my favorite female vocalists come from metal/symphonic metal bands though:
- Sharon den Adel (Within Temptation)
- Cristina Scabbia (Lacuna Coil)
- Angela Gossow (Arch Enemy)
- Liv (Leaves' Eyes)

My favorites: Lemuria, Gregory and the Hawk, Azure Ray, Warpaint, Bat for Lashes.
Not sure if that's the kind of thing you're looking for, but there it is.

I'll second anyone who mentioned Siouxie and the Banshees. Sharon Den Adel and Within Temptation are amazing. You might like Emilie Autumn's early stuff from the album Enchant, if you're into piano ballads and gorgeous violin melodies.

I saw that somebody already posted Cat Power. That's a great one :)
I also second Metric as well as the Yeah Yeah Yeahs, and I love Fiona Apple.
If you're into grunge female vocalists, I'd say check out Garbage, Jack Off Jill and Veruca Salt.
The XX has a female vocalist I like a lot.
Gregory and the Hawk
Oh, and I would highly recommend Angus and Julia Stone. It's the male vocalist more often than not, but the female vocalist has a really interesting voice. I think she made have done a solo project, but I can't remember right now.

If you are in the mood for a girl band you should definitly try The Like (brilliant 60s style LA based girl band who have amazing voices, lovely style and play their own instruments) or maybe Robots in Disguise (if you like a sort of electro-pop sound). Courtney Love and Hole have some great songs. Oh, and Beth Ditto from Gossip has great vocals.
For singers you should try famous 60s female vocalists like Janis Joplin, Grace Slick (from Jefferson Airplane/Starship), Joni Mitchell and Melanie (all of whom are fabulous). Also, Kate Bush (who has such an interesting and unique voice) and Stevie Nicks (from Fleetwood Mac). If you're more interested in modern music then you should try Emily Browning (an Australian actress, but she has a few songs, all of which are lovely), Skunk Anansie, Laura Marling, Kate Nash, Lily Allen etc. And if you like jazz, I reccomend Amy Winehouse or Imelda May (lovely Irish singer with two brilliant albums).

Paloma Faith
Lara Johnston
Gail Ann Dorsey
Linda Perry (4 Non Blondes, Deep Dark Robot, solo work)
Also, even though everyone knows 'em already, you really can't go wrong with Lady Gaga or Amy Winehouse... and if you're interested in older stuff, Etta James is where it's at.
